研究目的：

主要研究环泊酚致患儿意识消失的ED50，以及艾司氯胺酮对环泊酚致患儿意识消失ED50的影响  

Objectives of Study：

The main focus of this study is on the ED50 of consciousness loss in pediatric patients induced by cyclopofol, as well as the effect of esketamine on the ED50 of consciousness loss in pediatric patients induced by cyclopofol.

药物成份或治疗方案详述：

 

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.择期行全身麻醉手术的患儿 2.ASA分级I或II级; 3.年龄1-4岁; 4.性别不限； 5.体质量指数13~20kg/m2;

Inclusion criteria

1. Children undergoing elective general anesthesia surgery 2. ASA classification I or II; 3. Age range of 1-4 years old; 4. Gender is not limited 5. Body mass index 13-20kg/m2;

排除标准：

1.贫血、营养不良、肝肾功能异常者； 2.严重心脏血管疾病； 3.智力障碍者； 4.有神经精神系统疾病； 5.有麻醉药物过敏史； 6.服用镇静催眠类药物史；

Exclusion criteria：

1.Patients with anemia, malnutrition, or significant hepatic/renal dysfunction; 2.Severe cardiovascular diseases; 3.Intellectual disability; 4.Neuropsychiatric disorders; 5.History of hypersensitivity to anesthetic agents; 6.Chronic use of sedative-hypnotic medications within 30 days prior to enrollment.

环泊酚初始剂量为推荐的最佳剂量0.5mg/kg，30s内缓慢静脉注射完成给药。给药完成后每30秒进行一次警觉/镇静评估（OAA/S）评分，OAA/S评分≤1分则判定意识消失，3min内意识消失则为阳性指标。若患儿出现阳性指标，则下一例患者递减环泊酚剂量；若出现阴性指标，则下一例患儿递增环泊酚剂量。从出现阴性反应的前两例患者作为研究起点，直至出现7个阳性反应和阴性反应交叉点时结束研究。

Intervention：

The initial dose of cyclopofol was 0.5mg/kg, and the administration was completed by slow intravenous injection within 30s. The OAA/S score was performed every 30 seconds after the completion of administration, and the OAA/S score was determined to be disappearance of consciousness if ≤ score was 1, and the loss of consciousness within 3 minutes was a positive indicator. If the child has a positive index, the next patient will have a reduced dose of cyclopofol; If a negative indicator occurs, the next child will receive an escalating dose of cranpopol. The study starts with the first two patients with negative reactions and ends when there are 7 positive and negative intersections.
