研究目的：

本研究将建立基于CLIP-PCR的人布鲁氏菌的分子诊断方法，为病原菌检测提供一种免提核酸直接检测全血中RNA的新技术，以提高人布鲁氏病的诊断能力，实现布鲁氏菌病的早期识别和精准治疗。建立了灵敏高效的通用TaqMan探针法CLIP-PCR检测人感染布鲁氏 菌，成功应用于布鲁氏菌的血液样本检测，提高布鲁氏菌检测的敏感性，检测出更低拷贝的布鲁氏菌，为布鲁氏菌病的传播的控制、大规模筛查奠定了基础。  

Objectives of Study：

This study will establish a molecular diagnostic method for human Brucella based on CLIP-PCR, providing a new technology for pathogen detection that eliminates the need for nucleic acid extraction and directly detects RNA in whole blood. This will enhance the diagnostic ability for human brucellosis and enable early identification and precise treatment of brucellosis. A sensitive and efficient universal TaqMan probe method, CLIP-PCR, for detecting human Brucella infection has been established and successfully applied to the detection of Brucella in blood samples, improving the sensitivity of Brucella detection and enabling the detection of lower-copy Brucella. This lays the foundation for the control of Brucella transmission and large-scale screening.

纳入标准：

入选标准：年龄18岁以上；符合布鲁氏菌病诊断标准的疑似、临床诊断或确诊标准； 阴性对照：年龄18岁以上；无布鲁氏菌病流行病学史、不符合布鲁氏菌病症状且血培养未检测出布鲁氏菌；

Inclusion criteria

Inclusion criteria: age over 18 years old; Suspected, clinical or confirmed diagnostic criteria for brucellosis; Negative control: age over 18 years old; No epidemiological history of brucellosis, no symptoms of brucellosis, and no Brucella positive blood culture;

排除标准：

活动期恶性肿瘤、时期器官移植术后、造血干细胞移植术后、严重免疫系统疾病；使用免疫抑制药物、生物制剂或靶向药治疗者；拒绝签署知情同意书。

Exclusion criteria：

Active malignant tumors, organ transplantation after the period, hematopoietic stem cell transplantation after the period, severe immune system diseases; those who are treated with immunosuppressive drugs, biological agents or targeted drugs; those who refuse to sign the informed consent form.
