本研究旨在探索Omega-3脂肪酸在年龄相关性黄斑变性(AMD)中的关键作用,并揭示其对细胞线粒体自噬的调控机制,以阐明其在AMD发展中的影响,从而为Omega-3脂肪酸的临床应用提供理论依据。同时,通过深入解析AMD的发病机制,特别是抗氧化与抗炎干预策略的作用,本研究旨在为开发延缓AMD进展的新型治疗手段提供科学依据和潜在干预方向。  

Objectives of Study:

This study aims to investigate the critical role of omega-3 fatty acids in age-related macular degeneration (AMD) and elucidate their regulatory mechanisms on mitochondrial autophagy, providing mechanistic insights into how they influence AMD progression. By uncovering these pathways, we seek to establish a strong theoretical foundation for the clinical application of omega-3 fatty acids in AMD treatment. Furthermore, through an in-depth exploration of AMD pathogenesis—particularly focusing on antioxidant and anti-inflammatory interventions—this research aims to identify novel therapeutic strategies to effectively slow disease progression.

纳入标准:

1.符合AMD诊断标准(病例来自导师课题组:鱼籽油磷脂ω-3 视网膜病变临床观察研究课题);

Inclusion criteria

1.Diagnosed with AMD;

排除标准:

(1).研究眼存在不可控制的青光眼(定义为经规范治疗后眼压≥30 mmHg,或基于研究者在筛选或基线时期的判断);(2)存在活动性眼内、眼周的炎症或感染(例如角膜炎);(3)研究眼屈光介质严重浑浊,导致无法获取足够清晰的眼部影像学资料,如SD-OCT、FA、彩色眼底照相(CFP)等,影响研究者对安全性和疗效观察;(4)有遗传性出血倾向或凝血功能障碍;(5)3个月内有服用Omega-3类膳食补充剂者。(6)玻璃体手术史。以未发AMD的患者为对照组。

Exclusion criteria:

1.Uncontrolled glaucoma in the study eye (defined as intraocular pressure >= 30 mmHg despite standard treatment or based on investigator's judgment during screening/baseline). 2.Active intraocular or periocular inflammation/infection (e.g., keratitis). 3.Severe opacity of refractive media in the study eye precluding acquisition of adequate-quality ocular imaging (e.g., SD-OCT, FA, CFP), which may compromise safety/efficacy assessments. 4.Hereditary hemorrhagic tendency or coagulation disorders. 5.Use of Omega-3 dietary supplements within 3 months prior to enrollment. 6.History of vitreous surgery.

Standardized visual acuity testing (BCVA using ETDRS charts, LogMAR units), optical coherence tomography (OCT for retinal thickness measurement) with angiography (OCTA for capillary density analysis), microperimetry (retinal sensitivity assessment), multifocal electroretinography (mfERG per ISCEV standards), mitophagy-related proteins (PINK1/Parkin detection via Western blot), inflammatory markers (IL-6/TNF-α quantification by ELISA), and transcriptomic analysis.
