研究目的：

评价二甲双胍防治FAP息肉生长的疗效，探讨二甲双胍对FAP息肉发展的抑制作用和对Wnt/β-catenin信号通路的影响，旨在明确二甲双胍是否通过影响Wnt/β-catenin信号通路抑制肠道息肉的生长，从而抑制FAP的发展。  

Objectives of Study：

To evaluate the efficacy of metformin in the prevention and treatment of the growth of FAP polyps, and to explore the inhibitory effect of metformin on the development of FAP polyps and the effect on the Wnt/β-catenin signalling pathway, aiming to determine whether metformin inhibits the growth of intestinal polyps by affecting the Wnt/β-catenin signalling pathway, thereby inhibiting the development of FAP.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1.>=18 岁且符合 FAP 临床诊断标准的患者； 2.具备以下其中1条即可： （1）具有完整的结肠并伴有中度腺瘤负荷（100 至 1000 个腺瘤性息肉），并且目前正在考虑进行预防性手术的患者； （2）已行外科手术治疗，主要术式包括结肠次全切除术＋回肠肛管吻合术（ileo-rectal anastomoses，IRA）和回肠储袋-肛管吻合术（ileal-pouch-anal-anastomosis，IPAA）

Inclusion criteria

1. >=Patients aged 18 years who meet clinical diagnostic criteria for FAP; 2. One of the following is sufficient: (1) Patients with an intact colon with moderate adenomatous burden (100 to 1000 adenomatous polyps) and who are currently being considered for prophylactic surgery; (2) Surgical treatment has been performed, and the main surgical methods include subtotal colectomy + ileo-rectal anastomoses (IRA) and ileal-pouch-anal-anal-anal anastomosis (IPAA)

排除标准：

1.合并其他可能会影响肠道微生态的疾病，如炎症性肠病、肠易激综合征、短肠综合征引起的吸收不良； 2.纳入研究前 3 个月内服用过免疫抑制剂、糖皮质激素；纳入研究前 1 个月服用过抗生素、益生菌、益生元等；纳入研究前 2 周内服用过质子泵抑制剂、含铋剂药物等。 3.诊断为恶性疾病，如 CRC； 4.在试验前 6 个月内使用非甾体抗炎药每周 3 次以上； 5.诊断为糖尿病、怀孕或哺乳； 6.肝功能、肾功能检查结果显著异常，或有重大感染、呼吸道疾病等； 7.备孕的男性； 8.研究者认为存在有不合适入选因素的患者。

Exclusion criteria：

1. Combined with other diseases that may affect the intestinal microbiota, such as inflammatory bowel disease, irritable bowel syndrome, and malabsorption caused by short bowel syndrome; 2. Immunosuppressants, glucocorticoids within 3 months prior to inclusion in the study; Have taken antibiotics, probiotics, prebiotics, etc. 1 month before the inclusion study; Have taken proton pump inhibitors, bismuth-containing medications, etc., within 2 weeks prior to inclusion in the study. 3. Diagnosis of malignant disease, such as CRC; 4. Use of non-steroidal anti-inflammatory drugs more than 3 times per week within 6 months prior to the trial; 5. Diagnosed with diabetes, pregnancy or breastfeeding; 6. Significant abnormalities in liver function and kidney function tests, or major infections, respiratory diseases, etc.; 7. Men trying to conceive; 8. Patients who are considered by the investigator to have factors that are not suitable for inclusion.
