研究目的：

通过对健康人群及胃癌患者血浆中circRNA的鉴定及差异分析，进一步筛选对胃癌早诊潜在相关性的circRNA，寻找胃癌液体活检的生物标志物。  

Objectives of Study：

By conducting identification and differential analysis of circRNA in the plasma of healthy individuals and gastric cancer patients, followed by a screening for circRNA potentially associated with the early diagnosis of gastric cancer, we aimed to identify biomarkers suitable for liquid biopsy in gastric cancer.

纳入标准：

1. 入选本研究胃癌患者的合格受试者必须符合以下所有标准： （1）经病理证实为胃癌 （2）术前未接受任何抗癌治疗（包括放化疗、免疫治疗、靶向治疗等） （3）具有完整的临床、病理、随访资料 （4）无既往胃癌病史及治疗史 2.入选本研究健康人的合格受试者必须符合以下所有标准： （1）体检证实无疾病 （2）既往无肿瘤病史 （3）具有完整的临床资料

Inclusion criteria

1. Eligible subjects selected as patients with gastric cancer in this study must meet all the following criteria: (1) Gastric cancer confirmed by pathology. (2) Did not receive any anti-cancer treatment before operation (including radiotherapy and chemotherapy, immunotherapy, targeted therapy, etc.) (3) Have complete clinical, pathological and follow-up data. (4) No previous history of gastric cancer and treatment history. 2. Eligible subjects selected as healthy people in this study must meet all the following criteria: (1) Physical examination proves that there is no disease. (2) No previous history of tumor. (3) Have complete clinical data.

排除标准：

（1）术前接受放化疗或免疫治疗或靶向治疗 （2）临床信息缺失 （3）既往患有其他恶性肿瘤

Exclusion criteria：

(1) Preoperative chemotherapy or immunotherapy or targeted therapy. (2) Lack of clinical information. (3) Suffering from other malignant tumors in the past.

本研究采用实时荧光定量PCR法，检测、筛选、评估胃癌肿瘤标志物-环状核糖核酸（circRNA），使用的设备为分光光度计(Nanodrop)、荧光定量PCR检测仪(Q6) (Quantstudio 6 flex) 。

Index test:

In this study, Real-time Quantitative Polymerase Chain Reaction was used to detect, screen and evaluate circRNA, a tumor marker of gastric cancer. The equipment used was a spectrophotometer (Nanodrop) and a fluorescence quantitative PCR detector (Q6) (Quantstudio 6 flex).

首先在医院病历系统查询、筛选（根据纳入、排除标准）并记录所有适合入组人群。筛选合格的胃癌患者，取患者术前血常规/生化常规检查后剩余标本以备实验，在患者根治性术后、手术室病理科获取癌组织和癌旁组织标本。筛选合格的健康人群，在检验科取血液检验后剩余标本，进行血浆分离，冻存，以备实验。

Randomization Procedure (please state who generates the random number sequence and by what method)：

Firstly, the hospital medical record system is used to query and screen (according to the inclusion and exclusion criteria) and record all the people who are suitable for this research. Screening qualified patients with gastric cancer, taking the remaining samples after preoperative blood routine examination for the experiment, and obtaining cancerous tissue and adjacent tissue samples after radical surgery from pathology department of operating room. Screening qualified healthy people, taking the remaining samples after blood examination in the department of clinical laboratory, and separating the plasma and freezing it for the experiment.
