研究目的：

本研究采用随机对照研究的试验设计，旨在探究盐酸二甲双胍灌肠制剂对溃疡性结直肠炎患者治疗的作用，研究对象为浙江大学医学院附属第一医院正在接受药物治疗或即将开始接受药物治疗的轻中度活动期溃疡性结直肠炎患者。以活动期患者8周的内镜缓解结合组织学改善作为主要终点，探究二甲双胍对溃疡性结直肠炎的治疗效果及作用机制。  

Objectives of Study：

This study was designed as a randomized controlled trial to investigate the effect of metformin hydrochloride enema on ulcerative colitis. The subjects were patients with mild to moderate active stage ulcerative colitis who were receiving or were about to receive drug therapy in the First Affiliated Hospital of Zhejiang University School of Medicine. Endoscopic remission combined with histological improvement in active patients for 8 weeks was used as the primary endpoint to explore the therapeutic effect and mechanism of metformin in the treatment of ulcerative colitis.

纳入标准：

1: 年龄18-70周岁，性别不限 2: 根据内镜、组织学、影像学评估确诊的溃疡性结直肠炎（累及直肠或/且乙状结肠）受试者 3: 正在接受药物治疗的受试者 4: 自愿签署知情同意书

Inclusion criteria

1: Age 18-70 years old, gender is not limited 2: Subjects with confirmed ulcerative colitis(involving rectum and/or sigmoid) were evaluated by endoscopy, histology, and imaging 3: Subjects receiving medication 4: Patients voluntarily sign informed consent forms

排除标准：

1: 重度溃疡性结肠炎受试者 2: 用全身或直肠类固醇治疗 3: 已知对任何研究药物过敏 4: 肝肾功能障碍 5: 妊娠和哺乳期 6: 结直肠癌病史 7: 全部或部分结肠切除术史 8: 目前或以前二甲双胍治疗女性多囊卵巢综合征 9: 糖尿病受试者有乳酸酸中毒史，疾病状态与低氧血症相关的受试者，包括心肺功能不全 10: 肠道病原体粪便培养阳性，粪便卵和寄生虫检查阳性

Exclusion criteria：

1: Patients with severe ulcerative colitis 2: Patients treated with systemic or rectal steroids 3: Patients known to be allergic to any investigational drug 4: Patients with liver and kidney dysfunction 5: Pregnant and lactating patients 6: Patients with a history of colorectal cancer 7: Patients with a history of total or partial colectomy 8: Current or former metformin therapy for female subjects with polycystic ovary syndrome 9: Diabetic subjects with a history of lactic acidosis, and subjects with disease status associated with hypoxemia, including cardiopulmonary insufficiency 10: Patients with positive fecal cultures for intestinal pathogens and positive tests for fecal eggs and parasites
