本项目拟在病理确诊的脂肪肝患者中,观察非肥胖与肥胖脂肪肝患者MBOAT7 rs641738位点T等位基因频率的差异,分析该基因变异与肝组织脂肪变严重程度及脂代谢间的相关性,明确该基因变异与非肥胖脂肪肝发生发展的关系;构建MBOAT7点突变的肝细胞模型,探索该基因变异影响肝细胞脂质沉积的机制。本研究将进一步明确我国非肥胖脂肪肝的发病机制,为NAFLD临床分型和精准医疗奠定基础,最终提高NAFLD的诊治水平。 |

Objectives of Study: |
This project intends to observe the difference in the frequency of T allele at MBOAT7 rs641738 locus between non-obese and obese fatty liver patients with pathologically confirmed fatty liver disease, analyze the correlation between this gene variation and the severity of fatty degeneration in liver tissue and lipid metabolism, and clarify the relationship between this gene variation and the occurrence and development of non-obese fatty liver disease. The model of MBOAT7-point mutation in hepatocytes was constructed to explore the mechanism of the gene mutation affecting lipid deposition in hepatocytes. This study will further clarify the pathogenesis of non-obese fatty liver in China, lay a foundation for clinical typing and precision medicine of NAFLD, and ultimately improve the diagnosis and treatment level of NAFLD. |

排除标准: |
(1)缺乏必要的人口学指标、身体测量学指标、检验学指标等 (2)缺乏必要的既往史及用药史 (3)合并其他原因的肝脏疾病,例如HBV、HCV感染、自身免疫性肝病等其他疾病;大量饮酒史(酒精摄入男性>210g/天,女性>140g/天);药物引起的继发性肝脏脂肪变,如他莫昔芬、激素等 (4)合并其他严重疾病,可能影响身体营养状态,例如恶性肿瘤、严重心脏、肺脏、肾脏等器质性疾病 |

Exclusion criteria: |
(1) Lack of necessary demographic indicators, body measurement indicators, laboratory indicators, etc (2) Lack of necessary prior history and medication history (3) Liver disease with other causes, such as HBV, HCV infection, autoimmune liver disease and other diseases; A history of heavy alcohol consumption (alcohol intake > 210g/ day for men and > 140g/ day for women); Secondary hepatic steatosis caused by drugs, such as tamoxifen, hormones, etc (4) Combined with other serious diseases that may affect the nutritional status of the body, such as malignant tumors, serious organic diseases of the heart, lungs, kidneys, etc |
