研究目的：

通过对比冈上肌肌腱炎（Supraspinatus Tendonitis，ST）患者不同针刺手段治疗前后血清外泌体 micro RNA 表达的变化，筛选不同针刺手法针刺效应相关的关键 micro RNA。比较文献报道的 micro RNA 与本次研究发现的 micro RNA 的差异，以期探索不同针刺手法治疗 ST 的潜在调节机制。  

Objectives of Study：

Acupuncture has a long history of being used in Chinese medicine for the treatment of Supraspinatus Tendonitis. However, molecular biomarkers for diagnosis and prognosis of ST and its treatment are lacking. This study aimed to explore whether acupuncture could regulate differentially expressed exosomal miRNAs between patients with ST and healthy controls (HCs) and to identify diagnostic biomarkers that helped differentiate ST patients from HCs and identify prognostic biomarkers that helped to predict the effect of acupuncture.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

① 符合上述 ST 的诊断标准，并且为单侧发病。 ② 年龄：20-60 岁之间（包含 20、60 岁）。 ③ 就医前一月内未进行任何相关于 ST 的中西医治疗，并且未因其他原因服用镇痛药。 ④ 患者精神及认知功能正常，并能配合并完成治疗全过程，自愿加入且签署知情同意书。 ⑤ 能够接受不同针刺手法治疗，并且无相关禁忌症。

Inclusion criteria

① The diagnosis criteria of ST were met, and the disease was unilateral. ②Age: between 20 and 60 years old (including 20 and 60 years old). ③ No Chinese and western medicine treatment related to ST was performed within one month before medical treatment, and no analgesics were taken for other reasons. ④ The patient has normal mental and cognitive functions, can cooperate and complete the whole process of treatment, and voluntarily join and sign the informed consent. ⑤ Can be treated with different acupuncture techniques, and there are no related contraindications.

排除标准：

① 正在接受其他有可能会影响观察指标的治疗。 ② 有任何针刺治疗相关禁忌症；操作局部皮肤破损的患者。 ③ 妊娠或哺乳期妇女；既往有出血倾向或凝血功能障碍病史的患者。 ④ 合并心脑血管等疾病、处于偏瘫状态的患者；合并肝、肾、内分泌系统等患有严重原发性疾病的患者；继发于风湿性关节炎、类风湿性关节炎、糖尿病等全身系统性疾病的患者。 ⑤ 兼有肩部其他疾病，如骨折、脱位、肱二头肌长头肌腱炎等其他肩部软组织损伤等。

Exclusion criteria：

① Are receiving other treatments that may affect the outcome measures. ② There are any contraindications related to acupuncture treatment; Handling patients with local skin damage. ③Pregnant or lactating women; Patients with a history of bleeding or coagulopathy. ④ Patients with cardiovascular and cerebrovascular diseases and hemiplegia; Patients with severe primary diseases of liver, kidney, endocrine system, etc.; Secondary to rheumatoid arthritis, rheumatoid arthritis, diabetes and other systemic diseases. ⑤ With other shoulder diseases, such as fracture, dislocation, biceps long head tendinitis and other shoulder soft tissue injuries.

随机分配序列由一名独立统计学家使用统计学软件SAS9.1PLAN句子编辑生成，该统计学家来自于四川大学华西医学院循证医学中心，不参与该实验研究。符合纳入标准的患者通过随机和盲法分配到2个研究组（EA,PN）,确保每一例患者分配到2个组的几率一致。随机分组的序列号有专门指定的一名助教负责保管隐藏，所有研究人员在最后结果完成后才能知晓相关结果。分配到有针灸治疗的针灸治疗师将被直接告知分配的组别。所有患者在分配好组别后均会被告知所分组别名称和相关干预措施。

Randomization Procedure (please state who generates the random number sequence and by what method)：

The random allocation sequence will be produced by an independent statistician via the PLAN sentences of the statistical software SAS9.1, who works in the Evidence- Based Center of the Sichuan University and is not involved in the study. The eligible patients will be randomly and blindly allocated to the two study groups (EA,PN), ensuring that each patient had a consistent probability of being assigned to all four groups. The sequence numbers of random groups are kept hidden by a special teaching assistant, and all researchers will not know the relevant results until the final results are completed. Acupuncture therapists assigned to acupuncture treatments will be informed directly of the assigned group. All patients will be informed of the group name and relevant interventions after they are assigned to the group.
