研究目的:

本研究筛选与肥胖型肌少症相关的外泌体(MicroRNA 差异分析),并通过生信分析、全蛋白质组学检测等方法,筛选肥胖型肌少症生物标志物,进行差异蛋白的筛选与聚类,构建小RNA测序文库,基于illumina技术测序平台,进行单端测序,比较样本间差异蛋白阐明基因与肥胖型肌少症之间的关系。其次根据检测肥胖型肌少症患者与健康人群基因表达的差异结果,进行横断面研究进一步分析肥胖型肌少症中差异表达的MicroRNA与脂代谢、骨骼肌功能的相关性,以期为疾病的早期诊断及治疗提供新的策略。  

Objectives of Study:

In this study, we screened exosomes associated with obesity sarcopenia (MicroRNA differential analysis), and screened biomarkers of obesity sarcopenia by means of analysis and whole-proteomics, screened and clustered differential proteins, and constructed a small RNA sequencing library. Secondly, according to the difference of gene expression between obese sarcopenia patients and healthy people, a cross-sectional study was conducted to further analyze the correlation between differentially expressed MicroRNA, lipid metabolism and skeletal muscle function in obese sarcopenia, so as to provide a new strategy for early diagnosis and treatment of the disease.

纳入标准:

1.签署书面知情同意告知书;
2.肌少性肥胖诊断为肌少症+肥胖诊断标准。

Inclusion criteria

1. Sign a written informed consent notice;
2. The diagnosis of myopenia obesity is myopenia+obesity.

排除标准:

1.有糖尿病足及其他造成下肢活动障碍疾病;
2.甲状腺功能异常、库欣综合征、垂体功能异常等内分泌疾病;
3.各种急慢性感染及其他免疫性疾病,如终末期肾病、肝功能衰竭、心力衰竭、呼吸衰竭;或伴有恶性肿瘤、妊娠者;
4.不能配合完成步行速度、握力的测定;
5.服用抗骨质疏松药物、糖皮质激素、甲状腺激素、性激素等药物;
6.长期饮酒史,即饮酒时间超过5年,或乙醇量男性>140 g/周,女性>70 g/周。

Exclusion criteria:

1. Have diabetes foot and other diseases that cause lower limb movement disorder;
2. Endocrine diseases such as thyroid dysfunction, Cushing's syndrome, pituitary dysfunction;
3. Various acute and chronic infections and other immune diseases, such as end-stage renal disease, liver failure, heart failure and respiratory failure; Or with malignant tumor or pregnancy;
4. Unable to cooperate in the measurement of walking speed and grip strength;
5. Take anti-osteoporosis drugs, glucocorticoids, thyroid hormones, sex hormones and other drugs;
6. Long-term drinking history, that is, drinking for more than 5 years, or alcohol consumption>140 g/week for men and>70 g/week for women.

数据采集和管理(说明:数据采集和管理由两部分组成,一为病例记录表(Case Record Form, CRF),二为电子采集和管理系统(Electronic Data Capture, EDC),如ResMan即为一种基于互联网的EDC:

1.数据记录: 研究者必须保证数据记录的完整性和准确性。数据记录须用黑色水笔清晰地填写。严禁对错误的数值和/或文字进行涂改,应用单线划掉错误之处,在旁边填写正确的数值/文字,并签署修改者的姓名缩写和修改日期。 2.数据检查: 项目组质控人员或监查员需检查研究的进行是否遵循试验方案。 试验过程中要定期核查受试者的知情同意及筛选纳入情况;确认所有数据记录填写正确,并与原始资料一致;所有错误或遗漏均已改正或注明,经研究者签名并注明日期;受试者在研相关特殊情况应确认并记录;确认所有不良事件均应记录在案,严重不良事件在规定时间内作出报告、记录在案并及时处理等。 3.数据保密: 研究原始资料均属于非公开信息,应保密。研究者应保证在研究资料中受试者使用匿名,并遵循相关法律法规规定,对受试者的数据、病情及相关信息进行保密。

Data collection and Management (A standard data collection and management system include a CRF and an electronic data capture:

1. Data recording: The investigator must ensure the completeness and accuracy of the data recorded. Data records must be clearly filled in with black ink. Correction of incorrect values and/or words is strictly prohibited. Use a single line to cross out errors, fill in the correct values/words next to them, and sign the modifier's initials and date of modification. 2. Data check: Quality control personnel or inspectors of the project team shall check whether the study is carried out in accordance with the test plan. The informed consent and screening inclusion of subjects should be checked regularly during the test. Ensure that all data records are filled in correctly and consistent with the original data; All errors or omissions have been corrected or noted, signed and dated by the investigator; The subject's special situation during the study should be confirmed and recorded; Confirm that all adverse events should be recorded, and serious adverse events should be reported, recorded and handled in a timely manner within the specified time. 3. Data Confidentiality: The original research materials are non-public information and should be kept confidential.
