研究目的：

重症肺炎是全球发病率和病死率极高的感染性呼吸系统疾病。年龄、基础疾病和免疫状态是影响患者预后的重要因素，其中免疫紊乱是重症肺炎发生、发展的重要因素之一。但目前尚缺乏对宿主免疫系统反应异质性和免疫反应状态评估的指标。本研究基于单细胞测序和多组学技术，探寻重症肺炎患者与非肺炎患者肺泡盥洗液等样本的免疫细胞差异表达图谱和基因、代谢产物的差异表达，以期找到能够评估重症肺炎患者免疫状态和预测患者预后的标志物，为重症肺炎患者的早期临床治疗方法的调整提供参考依据。  

Objectives of Study：

Severe pneumonia is an infectious respiratory disease with a high global incidence and mortality rate. Age, underlying disease, and immune status are important factors affecting patient prognosis, with immune dysfunction being one of the key factors in the occurrence and development of severe pneumonia. However, there is currently a lack of indicators for evaluating the heterogeneity of host immune system responses and immune response status. Based on single-cell sequencing and multi-omics technologies, this study explores the differential expression patterns of immune cells and genes/metabolites in bronchoalveolar lavage samples of patients with severe pneumonia and non-pneumonia controls, in order to identify biomarkers that can evaluate immune status and predict patient prognosis, providing reference for adjusting early clinical treatment methods for patients with severe pneumonia.

纳入标准：

1: 入住ICU≥48小时 2: 社区获得性重症肺炎患者

Inclusion criteria

1: 24-48h after ICU admission 2: severe community-acquired pneumonia patients

排除标准：

1: 年龄＜18岁 2: 非社区获得性重症肺炎患者 3: 恶性肿瘤患者 4: 有免疫性疾病或使用免疫抑制剂患者 5: 妊娠期患者

Exclusion criteria：

1: Age<18 years old 2: Patients with severe non-community-acquired pneumonia 3: Patients with malignant tumors 4: Patients with immune disease or use of immunosuppressants 5: Patients during pregnancy
