研究目的：

本研究拟在前期卵泡体外培养和AO构建部分工作基础上，清除因病切除卵巢的肿瘤细胞，体外重建自体组织人工卵巢（AO）。皮质作为人卵泡供给源，脱细胞处理髓质作为ECM支架，自体腹膜和脂肪组织分离间充质干细细胞提取外泌体作为自体激活剂，对卵泡激活和分离技术、卵泡植入后发育支持等重要环节进行研究和优化，并探讨外泌体原始卵泡激活机制，为卵泡有效利用和AO重建技术临床应用提供可能，也为未来人口老龄化生育储备提供新方式，为再生医学提供新的发展方向。  

Objectives of Study：

In this study, based on the previous work of follicle culture in vitro and AO construction, the tumor cells of ovariectomized ovary due to disease were removed and the artificial ovary with autogenous tissue (AO) was reconstructed in vitro. The cortex is used as the source of human follicles, the acellular medulla is used as the ECM scaffold, the autologous peritoneum and adipose tissue are separated from mesenchymal dry cells and exosomes are extracted as autoactivators, and the important links such as follicle activation and separation technology, and the development support after implantation of follicles are studied and optimized, and the mechanism of protofollicle activation of exosomes is discussed. It provides the possibility for the effective utilization of follicles and the clinical application of AO reconstruction technology. It also provides a new way for the fertility reserve of aging population in the future and a new development direction for regenerative medicine.

年龄大于45岁或已绝经妇女
