研究目的：

本研究拟针对传统中药川芎嗪的有主要有效成分川芎嗪，开发能够有效缓解酒精戒断综合征，促进成瘾行为消退，且能够有效防止复饮的治疗药物。本项目的实施将有助于拓展传统中药及其主要有效成分川芎嗪的新功能（新适应症），可能将其发展成为一种有效的酒精成瘾治疗药物，并探究其神经影像机制。  

Objectives of Study：

This study intends to develop a therapeutic drug that can effectively relieve alcohol withdrawal syndrome, promote the regression of addictive behaviors, and effectively prevent re-drinking for the main active ingredient of traditional Chinese medicine ligustrazine. The implementation of this project will help to expand the new functions (new indications) of traditional Chinese medicine and its main active ingredient ligustrazine, which may be developed into an effective drug for the treatment of alcohol addiction, and explore its neuroimaging mechanism.

纳入标准：

病例入选标准 1）酒精成瘾： ①年龄18~65 岁； ②符合DSM-IV 酒精依赖的诊断标准； ③既往无明确神经系统疾病史，无精神疾病家族史； ④自愿参与研究，配好随访及观察，依从性好； ⑤无明显精神病性症状。 2）结合川芎嗪的治疗适应症，入选病例须满足动脉硬化指数≥4： 正常被试入组标准： ①年龄、性别与酒精成瘾病例匹配； ②无连续饮酒史，不符合任何物质依赖诊断，未使用过毒品； ③无任何严重神经系统或精神疾病；未经历脑外伤或其他脑组织受损情况； ④短期内无服用任何精神类药物； ⑤无认知功能障碍。

Inclusion criteria

Case Inclusion Criteria 1) Alcohol addiction: ①Aged 18~65 years old; ② Meet the diagnostic criteria of DSM-IV alcohol dependence; ③No clear history of neurological diseases in the past, no family history of mental diseases; ④Voluntary participation in research, with good follow-up and observation, and good compliance; ⑤ No obvious psychotic symptoms. 2) Combined with the treatment indications of ligustrazine, the selected cases must meet the arteriosclerosis index ≥ 4: Inclusion criteria for normal subjects: ① Age, gender, and alcohol addiction cases matched; ② No history of continuous drinking, no substance dependence diagnosis, no drug use; ③No serious nervous system or mental disease; no experience of traumatic brain injury or other brain tissue damage; ④ Not taking any psychotropic drugs in a short period of time; ⑤ No cognitive dysfunction.

排除标准：

病例排除标准 ①急性酒精戒断反应期，CIWA-Ar > 9 分； ②伴有除慢性酒精依赖外的其它疾病所导致的严重神经系统或精神疾病：如脑卒中、颅内感染、脑肿瘤、精神分裂症等； ③曾经经历脑外伤或其他脑组织受损情况； ④正在或短期内有服用其他任何精神类药物，使用毒品或存在其他物质依赖； ⑤存在川芎嗪应用禁忌症，或女性怀孕等不宜用药情况； ⑥存在不宜做头部磁共振检测情况，如幽闭恐惧症、体内有金属物品等。 正常被试排除标准： ①常规体检（体查、血液化验、心电图）提示有明显躯体疾病； ②存在不宜做头部磁共振检测情况，如幽闭恐惧症、体内有金属物品等。

Exclusion criteria：

Case Exclusion Criteria ① Acute alcohol withdrawal phase, CIWA-Ar > 9 points; ② Severe neurological or mental illnesses caused by diseases other than chronic alcohol dependence: such asStroke, intracranial infection, brain tumor, schizophrenia, etc.; ③ Have experienced a traumatic brain injury or other damage to brain tissue; ④ Taking any other psychotropic drugs, drug use, or other substance dependence in the short term; ⑤ There are contraindications to the application of ligustrazine, or women are pregnant and other conditions that are not suitable for drug use; ⑥ There are conditions that are not suitable for head MRI examination, such as claustrophobia, metal objects in the body, etc. Exclusion criteria for normal subjects: ①Routine physical examination (physical examination, blood test, electrocardiogram) shows prominent physical diseases; ② There are conditions that are not suitable for head MRI examination, such as claustrophobia, metal objects in the body, etc.

采用循环随机法——采用R软件中循环生成随机数（每个循环在1、2、3中形成随机性）对研究对象随机分组，1、2、3分别对应不同治疗组。随机数由非研究者操作生成。受试者按顺序入组，药物治疗也由非研究者的护士完成。研究者和被试均不知所分组别。

The cyclic random method is used to randomly generate random numbers in the R software (each cycle forms randomness in 1, 2, and 3) to randomly group the research subjects, and 1, 2, and 3 correspond to different treatment groups. Random numbers are generated by non-researcher operations. Subjects were enrolled sequentially, and drug treatment was also completed by a non-researcher nurse. Both the researchers and the subjects were unaware of the grouping.
