Objectives of Study：

To evaluate the efficacy and safety of SHR0410 injection for postoperative analgesia after laparoscopic surgery under elective general anesthesia.

药物成份或治疗方案详述：

 

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1. 在开始试验相关的活动前，自愿签署知情同意书，充分了解本次试验的目的和意义，并自愿遵守本试验流程;
2．择期在全身麻醉下行腹腔镜手术的受试者;预计手术持续时间≥1h;
3．美国麻醉医师协会(ASA)分级为I~II级;
4．年龄≥18 岁，性别不限;
5．18kg/m2≤体重指数(BMI)≤28kg/m2;
6．术后(从最后一针缝合完毕开始计时)4h 内，受试者任意时刻静息状态下 NRS≥4 分;
7．筛选期前 30 天至末次用药后 30 天内无捐精及生育计划，且受试者
及其伴侣均同意试验期间及末次用药后 30 天内采取经医学认可的
高效避孕措施(详见附件 9);
8．未绝经或绝经未满两年的女性受试者在研究入组前的 7 天内血妊
娠试验检查阴性，且未处于哺乳期。

Inclusion criteria

1. Before starting the trial-related activities, voluntarily sign the informed consent form, fully understand the purpose and significance of the trial, and voluntarily abide by the trial process;
2. Subjects who elect to undergo laparoscopic surgery under general anesthesia; the expected duration of surgery is ≥1h;
3. The American Society of Anesthesiologists (ASA) is graded I to II;
4. Age ≥ 18 years old, no gender restrictions;
5. 18kg/m2≤body mass index (BMI)≤28kg/m2;
6. Within 4 hours after the operation (starting from the completion of the last stitch), the subject's NRS at rest at any time is ≥ 4 points;
7. There were no plans to donate sperm or have a child 30 days before the screening period and 30 days after the last dose, and both subjects and their partners agreed to use medically approved highly effective contraception during the trial period and 30 days after the last dose (see Annex 9).
8. Female subjects who were not menopausal or had been menopausal for less than two years should have a negative blood pregnancy test within 7 days before study entry and were not lactating.

排除标准：

1. 筛选前 1 周内有头晕、恶心、干呕、呕吐或合并前庭功能紊乱;
2. 筛选前 6 个月内有新发心肌梗死或不稳定心绞痛病史，或有II度及II度以上房室传导阻滞等严重心律失常病史，或 NYHA 心功能III级及以上病史的受试者;
3. 诊断为难治性高血压或合并难治性高血压病史(详见附件 10);
4. 既往有脑卒中病史;
5. 既往有认知功能障碍，或有癫痫病史;
6. 既往有困难气道病史，如阻塞性睡眠呼吸暂停综合征等;
7. 合并混淆术后疼痛评价的其他身体疼痛情况;
8. 筛选期收缩压≤90mmHg(不包括从开始麻醉诱导到出恢复室期间的异常);
9. 筛选期丙氨酸氨基转氨酶(ALT)和/或天门冬氨酸氨基转氨酶(AST)>3.0 倍正常值上限，总胆红素(TBIL)>1.5 倍正常值上限，血肌酐(Cr)>1.5 倍正常值上限;
10. 筛选期静脉血钾(K+)<正常值下限，和/或静脉血钠(Na+)>正常

Exclusion criteria：

1. Dizziness, nausea, retching, vomiting or combined vestibular dysfunction within 1 week before screening;
2. Subjects with a history of new myocardial infarction or unstable angina, a history of severe arrhythmias such as atrioventricular block of grade II or higher, or a history of NYHA class III or higher heart function within 6 months before screening;
3. Diagnosed with refractory hypertension or a history of refractory hypertension (see Annex 10 for details);
4. Past history of stroke;
5. Cognitive dysfunction in the past, or a history of epilepsy;
6. History of difficult airway, such as obstructive sleep apnea syndrome;
7. Combined with other physical pain conditions that confound postoperative pain evaluation;
8. Systolic blood pressure ≤90mmHg during the screening period (excluding abnormalities during the period from the start of anesthesia induction to the recovery room);
9. Alanine aminotransferase (ALT) and/or aspartate aminotransferase (AST) >3.0 times the upper limit of normal, total bilirubin (TBIL) >1.5 times the upper limit of normal, serum creatinine (Cr) during the screening period >1.5 times the upper limit of normal;
10. Venous potassium (K+) normal during the screening period.
