研究目的：

1 明确中医药防治食管癌术后复发转移的疗效和安全性 2.探索食管鳞癌术后辅助治疗的优选方案，探索不同治疗的优势人群和适宜疗法； 3.探索食管鳞癌术后中医证型的变化规律及其与预后的相关性； 4.基于代谢组学技术探索食管鳞癌中医证型的生物学基础； 5.基于代谢组学探索食管鳞癌预后代谢分子/谱。  

Objectives of Study：

1.To determine the efficacy and safety of traditional Chinese medicine in the prevention and treatment of postoperative recurrence and metastasis of esophageal cancer; 2.To explore the optimal scheme of postoperative adjuvant therapy for esophageal squamous cell carcinoma, and to explore the dominant groups and appropriate treatments for different treatments; 3.To explore the changes of TCM syndrome types after operation of esophageal squamous cell carcinoma and its correlation with prognosis; 4.To explore the biological basis of TCM syndrome types of esophageal squamous cell carcinoma based on metabonomics; 5.Exploration of prognostic metabolic molecules / spectra of esophageal squamous cell carcinoma based on metabonomics.

药物成份或治疗方案详述：

 

Description for medicine or protocol of treatment in detail：

 

纳入标准：

（1）R0 术后病理分期 II-III 期淋巴结阳性食管鳞癌患者；诊断标准参考食管癌诊疗规范（2018版）；分期标准：采用 UICC/AJCC TNM 食管鳞癌分期系统（2017 年第 8 版）；
（2）术后完成化疗或放化疗 2 个月之内，或不接受或不耐受放化疗；经检查证实无复发转移；
（3）ECOG 评分≤2 分，且预计生存时间＞3 个月；
（4）年龄 18-75 岁，无心、脑、肾、肝、血液等系统严重疾病者；无合并其他肿瘤；
（5）同意参加本次研究并签署知情同意书。

Inclusion criteria

(1) Postoperative pathological staging II-III stage II-III lymph node positive esophageal squamous cell carcinoma patients; diagnostic criteria refer to the criteria for diagnosis and treatment of esophageal cancer (2018 version); staging criteria: using UICC/AJCCTNM esophageal squamous cell carcinoma staging system (8th edition 2017);
(2) Postoperative chemotherapy or radiotherapy within 2 months, or do not accept or do not tolerate radiotherapy and chemotherapy; confirmed by examination that there is no recurrence and metastasis;
(3) ECOG score ≤ 2, and expected survival time > 3 months;
(4) 18-75 years old, no serious diseases of heart, brain, kidney, liver, blood and other systems; no other tumors;
(5) Agreed to participate in this study and signed the informed consent form.

排除标准：

（1）孕妇或哺乳期妇女；
（2）已知对研究用药过敏的患者；
（3）正在进行其他药物试验的患者；
（4）合并有心、肝、肾、心脏等主要脏器存在非肿瘤性严重疾病和功能障碍、活动性脑血管病变、不受控制的感染和代谢性疾病；
（5）研究者判定不适合参加本研究。

Exclusion criteria：

(1) Pregnant or lactating women;
(2) Patients who are known to be allergic to research drugs;
(3) Patients who are conducting other drug trials;
(4) Severe non-neoplastic diseases and dysfunction in major organs such as heart, liver, kidney and heart, active cerebrovascular disease, uncontrolled infection and metabolic diseases;
(5) The researchers determined that it was not suitable to participate in this study.

本研究采用双盲设计。试验设计者（不参与试验实施）负责药物编盲过程并保存盲底。在外观完全一致的中药颗粒剂和安慰剂包装盒上标以携带随机信息的药物编号。研究者按纳排标准招募筛选受试对象，根据受试对象的随机分组情况发放药物，并完成随访工作。直到试验 完成时，受试对象、研究者和统计分析师对受试对象入组和治疗情况处于盲态。

Blinding：

Double-blind design was adopted in this study. The trial designer (not involved in the implementation of the trial) is responsible for the drug blindness process and preserving the blind bottom. The packing boxes of traditional Chinese medicine granules and placebos that are identical in appearance are marked with the drug number carrying random information. The researchers recruited and screened the subjects according to the admission criteria, distributed drugs according to the random grouping of the subjects, and completed the follow-up work. Until the completion of the experiment, the subjects, researchers and statistical analysts were blind to the enrollment and treatment of the subjects.
