本项目拟系统全面研究MANF蛋白与脓毒症肺损伤疾病进展的关系，探明MANF在脓毒症肺损伤中的分子诊断价值；并以内质网应激介导细胞凋亡为核心，研究重组人MANF对LPS脓毒症小鼠肺损伤转归的影响及分子机制，为开拓MANF作为早期诊断、治疗脓毒症肺损伤的新途径提供了很好的思路和方法。  

Objectives of Study：

This project intends to systematically and comprehensively study the relationship between MANF protein and the progression of septic lung injury, and to explore the molecular diagnostic value of MANF in septic lung injury. The effect and molecular mechanism of human MANF on the outcome of lung injury in LPS mice with sepsis provides a good idea and method for developing MANF as a new way of early diagnosis and treatment of septic lung injury.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

脓毒症组标准为：当患者存在感染或疑似感染，其脓毒症相关序贯器官衰竭评分较基线上升（sequential organ-failure assessment，SOFA）≥2。或至少满足床旁快速SOFA（quick SOFA, qSOFA）中的两项时可诊断为脓毒症。对照组为满足SIRS但无脓毒症患者。其标准为：体温≥38℃或≤36℃，心率≥90次/分钟，呼吸速率≥20/min或过度通气PaCO2≤32 mmHg，白细胞增多(≥12,000/cu mm)或白细胞减少(≤4000/cu mm)。

Inclusion criteria

The criteria for the sepsis group were as follows: when patients had infection or suspected infection, their sepsis-related sequential organ-failure assessment (SOFA) score was ≥2 from baseline. Or at least two of the bedside rapid SOFA (quick SOFA, qSOFA) criteria can be diagnosed as sepsis. The control group consisted of patients who met SIRS but did not have sepsis. The criteria are: body temperature ≥38°C or ≤36°C, heart rate ≥90 beats/min, respiratory rate ≥20/min or hyperventilation PaCO2 ≤32 mmHg, leukocytosis (≥12,000/cu mm) or leukopenia (≤4000/ cu mm).

排除标准：

年龄小于18岁的患者及其他非SIRS或脓毒症的患者。
