研究目的：

ARID1A 在直肠癌中突变率可达到20-40%。因此了解ARID1A分子功能并确定是否可以通过药物靶向治疗ARID1A突变的直肠癌患者，具有重要的临床前景。目前研究发现ARID1A 的缺失会损害 DNA 损伤修复，而诱导的 DNA 损伤负担可能会增加癌细胞对ATR 依赖性 DNA 修复途径的依赖，以维持基因组完整性。所以，ARID1A突变细胞可能对 ATR抑制剂治疗敏感。通过肿瘤体外药敏试验三磷酸腺苷生物发光法（ATP-TCA）检测技术对比ARID1A阳性及ARID1A阴性直肠癌细胞对ATR抑制剂药物敏感。  

Objectives of Study：

The mutation rate of ARID1A in colon cancer can reach 20-40%. Therefore, understanding ARID1A molecular function and determining whether patients with rectal cancer with ARID1A mutations can be treated by drug targeting holds significant clinical promise. Current studies have found that ARID1A deletion impairs DNA damage repair, and the induced DNA damage burden may increase cancer cells' reliance on ATR-dependent DNA repair pathways to maintain genomic integrity. Therefore, ARID1A mutant cells may be sensitive to ATR inhibitor treatment. Comparison of ARID1A-positive and ARID1A-negative colon cancer cells sensitive to ATR inhibitor drugs by tumor in vitro drug sensitivity assay adenosine triphosphate bioluminescence (ATP-TCA) assay technique.

纳入标准：

于 2022 年 5 月至 2025 年 6 月在绵阳富临医院行原发性直肠癌肿瘤切除术的患者。

Inclusion criteria

Patients who underwent resection of primary colon cancer tumors at Mianyang Fulin Hospital from May 2022 to July 2025.

排除标准：

1: 接受术前化疗或术前放疗; 2不愿意参加这项研究的患者

Exclusion criteria：

1: Patients who received preoperative chemotherapy or preoperative radiation therapy; 2 Patients who did not want to participate in this study
