本研究通过双向凝胶电泳技术(2-DE)、基质辅助激光解析电离飞行时间串联质谱技术(MALDI-TOF-MS/MS)与非靶向代谢组学液相色谱-质谱技术,对急性心肌梗死患者的罪犯血栓斑块蛋白质组及血清代谢特征进行系统分析,筛选与急性心肌梗死的罪犯斑块分型相关的差异蛋白及代谢物,并与AMI患者临床特征改变相关联,筛选和验证灵敏度高、专属性强的AMI临床罪犯斑块分型血清代谢标志物,优化和构建准确率高、适用性广、方便快捷的诊断模型,为AMI的早期诊断鉴别、精准治疗提供科学依据。 |

Objectives of Study: |
In this study, two-dimensional gel electrophoresis, matrix-assisted laser desorption ionization time-of-flight tandem mass spectrometry, and non-targeted metabolomics liquid chromatography-mass spectrometry were used to analyze the proteomic and serum metabolic characteristics of culprit thrombus plaques in patients with acute myocardial infarction. Systematic analysis, screening of differential proteins and metabolites related to the culprit plaque typing of acute myocardial infarction, and correlation with changes in clinical characteristics of AMI patients, screening and validation of AMI clinical culprit plaque typing serum with high sensitivity and specificity Metabolic markers, optimizing and constructing a diagnostic model with high accuracy, wide applicability, convenience and rapidity, provide a scientific basis for the early diagnosis, identification and precise treatment of AMI. |

This study included patients with acute myocardial infarction who met the inclusion criteria and signed informed consent. According to the characteristics of the culprit lesions, the patients were divided into plaque rupture group and plaque erosion group. Therefore, randomization was not performed in this study. |
