研究目的：

首先，在原发性失眠发生的神经机制方面，借助多模态神经影像技术，多层面探索原发性失眠中枢神经系统脑结构及功能改变；其次，在原发性失眠干预治疗机制方面，纵向采集干预治疗前后的多模态神经影像数据，结合发生机制的神经证据，建构针灸干预治疗原发性失眠的有效指标，从客观的神经证据角度确定临床治疗的意义；最后，联合原发性失眠发生及干预治疗机制，明确原发性失眠的影响因素，有效指导临床诊断及治疗。  

Objectives of Study：

Firstly, in terms of the neural mechanism of primary insomnia, multi-modal neuroimaging technology was used to explore the brain structure and function of the central nervous system of primary insomnia. Secondly, in terms of intervention mechanism for primary insomnia, multi-modal neuroimage data before and after intervention were collected longitudinally, combined with neurological evidence of pathogenesis, effective indicators of acupuncture intervention for primary insomnia were constructed, and clinical significance was determined from the perspective of objective neurological evidence. Finally, combined with the occurrence and intervention mechanism of primary insomnia, the influencing factors of primary insomnia were determined to effectively guide clinical diagnosis and treatment.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

1. 主诉为入睡困难或难以维持睡眠,或睡眠质量差；这种睡眠紊乱每周至少发生3次并持续1个月以上；
2. 日夜关注于失眠,过分担心失眠的后果；
3. 睡眠量和(或)质的不满意引起了明显的苦恼或影响了社会及职业功能；
4. 体内无金属植入物, 无 MRI 扫描禁忌；
5. 试验前，通过针刺应答预试验筛除无针刺反应者和过度针刺反应者，即所有受试者接受针刺后均有针感。

Inclusion criteria

a. The chief complaint is difficulty falling asleep or maintaining sleep, or poor sleep quality; The sleep disorder occurs at least three times a week for more than a month;
b. Focus on insomnia day and night, worry too much about its consequences;
c. Dissatisfaction with the quantity and/or quality of sleep causes significant distress or affects social and occupational functioning;
d. No metal implants in the body and no contraindications of MRI scanning;
e. Prior to the test, those without acupuncture response and those with excessive acupuncture response were screened out through the preliminary test of acupuncture response, that is, all subjects had needle sensation after receiving acupuncture.

排除标准：

1. 失眠症状由药物、酒精引发，脑外伤史，进行过脑部手术；
b. 患者本人有精神或神经系统病史，入院时满足 Structured Clinical Interview for DSM-IV Axis I Disorders Diagnostic and Statistical Manual of Mental Disorders, 4th Edition Axis I Disorders (SCID I) 检查存在精神心理问题，一级亲属有精神病史，近三个月内服用过精神类药物；
d. 存在酒精依赖或药物滥用史。

Exclusion criteria：

a. Insomnia symptoms caused by drugs and alcohol, a history of brain trauma,had brain surgery;
b. The patient has a history of psychiatric or neurological problems. Meet the requirements of Structured Clinical Interview for DSM-IV Axis I Disorders Diagnostic and Statistical Manual of Mental Disorders With 4th Edition Axis I Disorders (SCID I), a first-degree relative has a history of mental illness or has taken psychotropic drugs in the last 3 months;
d. History of alcohol dependence or drug abuse.
