研究目的：

我们将收集威胁视力的糖尿病视网膜病变患者和健康成年人的粪便样本，并基于16S核糖体RNA (rRNA)基因测序描述了DR患者的人类肠道微生物群落结构，此外，我们系统地检测和识别DR和HC患者的肠道菌群组成和功能的差异。本研究旨在为糖尿病视网膜病变的潜在微生物靶点的诊断和识别提供一种非侵入性的方法，从而使糖尿病视网膜病变的发病机制和治疗策略的进一步研究成为可能。  

Objectives of Study：

We will collect stool samples from the patients with sight-threatening diabetic retinopathy and healthy individuals (HC) and delineate the community structure of the human gut microbiome of DR patients based on 16S ribosomal RNA (rRNA) gene sequencingin additionwe systematically detect and identify differences in the composition and function of the gut microbiota between patients with DR and HC. The study aimed to provide a non-invasive approache for diagnosis and identification of potential microbial targets that would enable further studies on the pathogenesis and therapeutic strategies for diabetic retinopathy.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

实验组的纳入标准为:(1)年龄25-70岁，(2)符合1999年世界卫生组织(WHO)诊断T2DM的标准，(3)根据国际临床糖尿病视网膜病变疾病严重程度量表(2002)诊断糖尿病视网膜病变。无论严重程度如何，视网膜病变患者均被分为DR组，(4)是否提供知情同意，(5)除DRIn外，无其他眼内疾病。正常受试者包括:(1)所有健康个体均为本项目糖尿病视网膜病变患者的配偶，与配偶共同生活，(2)健康个体与配偶的年龄差距不超过3岁;(3)除屈光不正外，无其他眼病，无糖尿病等系统性重大疾病;

Inclusion criteria

The inclusion criteria of the experimental group were as follows (1) age 25–70 years, (2) meeting the 1999 World Health Organization (WHO) diagnostic criteria for T2DM（3）Diabetic retinopathy was diagnosed based on the International Clinical Diabetic Retinopathy Disease Severity Scale (2002).Regardless of the level of severity,Patients with retinopathy, were classified into DR group, and(4) provision of informed consent，（5）They did not have any other intraocular diseases other than DRIn contrast,Normal subjects were included as follows:（1）All of the healthy individuals are the spouses of patients with diabetic retinopathy included in this project, who live with their spouses, and have no obvious dietary structure differences with their spouses;（2）The age difference between healthy individuals and their spouses is not more than 3 years;（3）They had no eye diseases other than refractive errors，no diabetes or ang other systemic major diseases；

排除标准：

排除标准如下:(1)纳入前6周内使用过抗生素者;(2)在粪便取样前3个月内使用益生菌和益生元;(3)腹部手术史;(4)每日饮酒>30 g;(5)严重免疫缺陷、严重肾病或肝病者;(6)哺乳或怀孕。

Exclusion criteria：

Exclusion criteria were the following:(1) those who had usage of antibiotics within 6 weeks before inclusion; (2) use of probiotics and prebiotics within 3 months prior to fecal sampling.; (3) history of abdominal surgeries（4）daily alcohol consumption >30 g; (5) significant immunodeficiencyserious kidney disease or iver disease ; and (6) breastfeeding or pregnancy.
