研究目的：

通过基因芯片技术结合生物信息学分析等方法，比较非综合征型单纯腭裂患者患侧腭组织和正常腭组织的基因表达情况，寻找差异表达的miRNA，并通过多对腭裂患侧组织及正常腭组织的PCR检测验证靶miRNA在腭裂组织与正常健康腭组织间的差异表达；原代培养正常人腭组织，并进一步通过转染技术构建靶基因上下调细胞模型，结合细胞增殖实验、伤口愈合实验、细胞周期与凋亡分析等验证靶miRNA对腭成纤维细胞功能的影响；构建腭裂缺陷动物模型验证靶miRNA在腭裂组织与正常健康组织间的差异表达。为非综合征型腭裂畸形早期的分子诊断和靶向治疗提供新的思路。  

Objectives of Study：

Through gene chip technology combined with bioinformatics analysis and other methods, the gene expression of the affected palate tissue and the normal palate tissue of patients with non-syndromic simple cleft palate were compared, and differentially expressed miRNAs were found. Tissue PCR detection verifies the differential expression of target miRNA between cleft palate tissue and normal healthy palate tissue; normal human palate tissue was cultured in primary culture, and a cell model of target gene up-regulation was further constructed by transfection technology, combined with cell proliferation experiments and wound healing experiments , cell cycle and apoptosis analysis to verify the effect of target miRNAs on the function of palate fibroblasts; construct cleft palate defect animal models to verify the differential expression of target miRNAs between cleft palate tissue and normal healthy tissue. To provide new ideas for early molecular diagnosis and targeted therapy of non-syndromic cleft palate deformity.

Description for medicine or protocol of treatment in detail：

 

纳入标准：

正常腭组织取样纳入标准：①患者无唇腭裂、无口腔癌症肿瘤等，腭组织健康，如为外伤患者需清创后切取组织；②患者年龄≤20岁；③取样不影响后期修复；④患者及其家属知情同意，通过医院伦理审查。
腭裂患者取样纳入标准：①患者仅患有非综合征型腭裂，无口腔癌症肿瘤等其他疾病；②患者年龄6-60个月；③取样不影响后期修复；④患者及其家属知情同意，通过医院伦理审查。

Inclusion criteria

Inclusion criteria for normal palate tissue sampling: ① The patient has no cleft lip and palate, no oral cancer tumors, etc., and the palate tissue is healthy. If it is a trauma patient, the tissue should be removed after debridement; ② The patient's age is ≤20 years old; ③ The sampling does not affect the later repair; ④ The patient and their families gave informed consent and passed the hospital ethics review.
Inclusion criteria for sampling of patients with cleft palate: ① The patient has only non-syndromic cleft palate-only and no other diseases such as oral cancer; ② The age of the patient is 6-60 months; ③ The sampling does not affect the later repair; ④ Informed consent of the patient and his family , passed the hospital ethics review.

排除标准：

正常腭组织取样排除标准：①患者患有唇腭裂、口腔癌症肿瘤等疾病，腭组织不健康；②患者年龄＞20岁；③取样影响后期修复；④患者及其家属知情不同意。
腭裂患者取样排除标准：①患者不患有非综合征型腭裂、或患有口腔癌症肿瘤等疾病；②患者年龄＞60个月；③取样影响后期修复；④患者及其家属知情不同意。

Exclusion criteria：

The exclusion criteria for normal palate tissue sampling: ① The patient suffered from cleft lip and palate, oral cancer tumors and other diseases, and the palate tissue was unhealthy; ② The age of the patient was more than 20 years old; ③ The sampling affected the later repair; ④ The patient and his family did not give informed consent.
Sampling exclusion criteria for patients with cleft palate: ① The patients did not suffer from non-syndromic cleft palate, or oral cancer and other diseases; ② The age of the patients was more than 60 months; ③ The sampling affected the later repair; ④ The patients and their families did not give informed consent.
