To investigate the intestinal microflora and the metabolite profiling changes in healthy population living at high altitude or plain, and to screen the key microbiota and target factors for predicting or affecting type 2 diabetes development and associated metabolic pathways. |

药物成份或治疗方案详述: |
利用代谢组学研究技术,检测高原和平原地区正常糖耐量受试者肠道菌群和血清代谢物质谱,分析筛选差异性菌群和表达代谢物。利用MetaboAnalyst分析软件和KEGG数据库进行差异代谢物代谢通路分析和互作网络构建。 1.研究对象和分组:纳入符合入组标准的受试者共51例。 2.一般资料收集:年龄、性别、种族、居住地年限、糖尿病病程/用药、高血压病史/用药、血脂异常/用药,心脑血管疾病史,吸烟史、饮酒史、抗血小板药物应用. 3.体检:身高、体重、腰围、臀围,计算体质指数(BMI);血压(SBP/DBP)、心率;血氧饱和度。 4.实验室检查: ① 留取血液标本测:红细胞计数(RBC ),血红蛋白(Hb),红细胞压积(Hct);空腹/餐后2小时血糖(FPG/P2hPG),空腹/餐后2小时胰岛素水平(FINS/P2hINS)和C肽水平;糖化血红蛋白(HbA1c);血尿酸(UA),血肌酐(Scr),血脂谱(TC, HDL-c, LDL-c, TG);超敏C-反应蛋白(hs-CRP);游离脂肪酸(FFA); ② 留取尿液测:尿微量白蛋白(ALB); ③ 计算:HOMA-IR=FPG(mmol/L)×FINS(mIU/L)/22.5; HOMA-β=20×FINS(mIU/L)/[FPG(mmol/L)-3.5] 5.留取血标本,离心留取血清后分装并保存于-80℃冰箱待送代谢组学检查(上海Biotree公司) ① 标本准备及预处理; ② 标本经色谱分离后进行质谱分析; ③ 数据采集与分析:PCA分析,PLS-DA分析,OPLS-DA分析,差异物质筛选; ④ 利用MetaboAnalyst分析软件和KEGG数据库进行差异代谢物代谢通路分析和互作网络构建。 6.留取粪便样本,液氮处理后分装并保存于-80℃冰箱待送16S和宏基因组测序(上海Biotree公司) ① 标本准备及预处理; ② 标本经DNA提取、质控等后进行测序分析; ③ 数据采集与分析:物种组成及丰度分析,基因功能及丰度分析,差异菌株筛选等。 |

Description for medicine or protocol of treatment in detail: |
Using metabolomics approaches, to detect alterations of serum metabolite profiling in individuals with normal glucose tolerance (NGT), impaired glucose tolerance (IGT), or type 2 diabetes (T2DM), which living at Tibet plateau or plain. Relevant metabolic pathway analysis will be performed by MetaboAnalyst software and KEGG pathway database. 1.Total of 180 subjects will be enrolled in this study and divided into higher altitude group (group H, altitude height >= 2800m) and lower altitude group (group L, altitude height <500m). Of each group, the subjects will be further divided into 3 subgroups (group NGT, group IGT, or group T2DM) based on their glucose tolerance status, 30 participants in each subgroup. 2.Clinical characteristics of the subjects will be collected such as age, gender, race, duration of residence, duration of diabetes, hypertension, or dyslipidemia, and etc. 3.Anthropometric measurements will also be collected such as height, body weight, waist circumference, hip circumference, systolic / diastolic blood pressure, heart beat, blood oxygen saturation (SaO2). 4.Lab examination: Blood samples will be collected for the detection of RBC, Hb, Hct, FPG/P2hPG, FINS/P2hINS, F-CP/P-2hCP, HbA1c, lipid profiles, and etc. HOMA-IR=FPG(mmol/L) x FINS(mIU/L)/22.5, HOMA-β=20 x FINS(mIU/L)/[FPG(mmol/L)-3.5] 5.Serum samples will be collected and stored at -80 degrees C for detection of metabolite profiling and analysis of metabolic pathways. 6.Feces samples will be collected,liquid nitrogen treated and stored at -80℃ for gut microbiota detection. |

常驻于高原或平原地区的汉族或藏族正常糖耐量者,年龄在40~80岁之间。入组前签署临床研究知情同意书。 |

Chinese Han or Tibetan residents living at Tibet plateau or plain with normal glucose tolerance (NGT) with age from 40~80 years. |
