研究目的：

主要目的：研究失眠障碍患者功能影像学、遗传学特征，探究失眠患者功能影像学特异性变化，尤其是PVT所在的中线丘脑区域,其兴奋性的改变及白质纤维连接特性变化，探索新的睡眠调控机制，为疾病的临床识别、诊断、治疗提供科学指标。  

Objectives of Study：

Main purpose: To study the functional imaging and genetic characteristics of patients with insomnia, to investigate the specific changes of functional imaging in patients with insomnia, especially the changes of excitability and white matter fiber connection characteristics in midline thalamic region where PVT is located,and explore new sleep regulation mechanisms, so as to provide scientific indicators for clinical identification, diagnosis and treatment of diseases.

纳入标准：

失眠障碍组：
1.右利手；
2.年龄18-60岁；
3.小学或以上文化程度，知晓汉语并能理解量表内容，完成量表者；
4.愿意加入该临床实验，能配合课题研究，签署知情同意书；
5.符合美国精神疾病诊断和统计手册第五版（DSM-5）中失眠障碍诊断标准；
6.匹兹堡睡眠质量指数量表（PSQI）≥5分；
7.符合入睡困难/睡眠维持困难/早醒的PSG表现，AHI<5次/时。
健康对照组：
1.右利手；
2.年龄18-60岁 ；
3.小学或以上文化程度，知晓汉语并能理解量表内容，完成量表者；
4.愿意加入该临床实验，能配合课题研究，签署知情同意书；
5.睡眠质量、睡眠习惯良好，无入睡困难、睡眠维持困难或早醒等睡眠问题；
6.PSQI≤4分，SAS标准分≤50分，SDS标准分≤53分；
7.符合该年龄段相适应的睡眠结构及各期睡眠比例的PSG表现，AHI<5次/时。

Inclusion criteria

Insomnia disorder group:
1. Right-handed;
2. Age 18-60 years old;
3. Primary school or above, those who know Chinese and understand the contents of the scale, who have completed the scale;
4. Willing to join the clinical trial, can cooperate with the subject research, and sign the informed consent;
5. Meet the diagnostic criteria for insomnia disorders in the American Diagnostic and Statistical Manual of Mental Disorders Fifth Edition (DSM-5);
6. Pittsburgh Sleep Quality Index (PSQI) >= 5 points;
7. In line with PSG performance of difficulty falling asleep/difficulty maintaining sleep/early awakening, AHI<5 times/hour.
Healthy control group:
1. Right-handed;
2. Age 18-60 years old;
3. Primary school or above, those who know Chinese and understand the contents of the scale, who have completed the scale;
4. Willing to join the clinical trial, can cooperate with the subject research, and sign the informed consent;
5. Good sleep quality, good sleep habits, no sleep problems such as difficulty falling asleep, difficulty maintaining sleep or waking up early;
6. PSQI<=4 points, SAS standard score<=50 points, SDS standard score<=53 points;
7. PSG performance in line with the age-appropriate sleep structure and the proportion of sleep in each stage, AHI<5 times/hour.

排除标准：

1.既往或目前患重大脑器质性疾病、严重心、肝、肾、肺及其他躯体疾病；
2.有磁共振禁忌症者，如植入血管支架、起搏器、金属假牙、助听器等，有幽闭恐惧症等；或磁共振发现大脑结构异常；
3.近2周服用抗焦虑、抑郁及睡眠药物史；
4.既往或目前物质滥用或依赖史；
5.患有其他重性精神疾病或家族史（如精神分裂症、双相情感障碍、精神发育迟滞等）；
6.女性在妊娠、哺乳期；或准备怀孕者。

Exclusion criteria：

1. Past or current suffering from severe organic brain disease, severe heart, liver, kidney, lung and other physical diseases;
2. Those with contraindications to magnetic resonance imaging, such as implantation of vascular stents, pacemakers, metal dentures, hearing aids, etc., claustrophobia, etc.; or abnormal brain structure found by magnetic resonance imaging;
3. History of taking anti-anxiety, depression and sleep drugs in the past 2 weeks;
4. Past or current history of substance abuse or dependence;
5. Suffering from other serious mental illness or family history (such as schizophrenia, bipolar disorder, mental retardation, etc.);
6. Women who are pregnant or breastfeeding; or who are planning to become pregnant.
