研究目的：

本项目从临床研究和动物实验2个层面，对小檗碱治疗胰岛素抵抗型多囊卵巢综合征（PCOS-IR）后，相关症状、免疫炎症、肠道菌群改善及PI3K-Akt信号通路蛋白的上下游控制关系进行研究，解析小檗碱改善肠道菌群、调节炎症反应、增加胰岛素敏感性的分子机制，为PCOS-IR治疗提供新的可筛选药物及研究方向。  

Objectives of Study：

This project from the clinical and animal experiments on the two aspects of berberine treatment of insulin resistance polycystic ovary syndrome (PCOS-IR), including related symptoms, immune inflammation, Gut microbiota, is to study that the upstream and downstream control relationship of the protein of PI3K-Akt signaling pathway, which is aimed to analyze the molecular mechanism of berberine to improve Gut microbiota, regulate inflammation and increase insulin sensitivity, and provide new screening drugs and research directions for PCOS-IR treatment.

纳入标准：

I、PCOS-IR 及 PCOS-NIR 患者：
①符合 PCOS 的鹿特丹诊断标准；
②采用 HOMA-IR 评价，HOMA-IR=空腹胰岛素（FINS）×空腹血糖（FPG）/22.5.≥2.69 为胰岛素抵抗。
③年龄在 18～40 岁之间的女性；
④本次治疗期间未曾使用过其他西药治疗；
⑤知情同意，自愿受试，并可追踪观察者。
II、正常人群：（选自我院体检中心）
①已建立正常月经周期，且有规律月经者；
②年龄在 18～40 岁之间的健康女性；
③知情同意，自愿受试，并可追踪观察者。

Inclusion criteria

PCOS-IR and PCOS-NIR patients:
1. Rotterdam diagnostic criteria in line with PCOS;
2. HOMA-IR evaluation, HOMA-IR = fasting insulin (FINS)*fasting blood glucose (FPG)/22.5 ≥ 2.69 for insulin resistance;
3. Women aged 18 to 40 years;
4. Informed consent, voluntary test, and can track observers.

Normal population: (selected from our medical examination center):
1. A normal menstrual cycle has been established, and regular menstruation;
2. Healthy women aged 18 and 40 years;
3. Informed consent, voluntary test, and follow-up observers.

排除标准：

①妊娠或哺乳期妇女；
②合并有心脑血管、肝肾和造血系统等严重原发性疾病、精神病患者；
③合并有恶性肿瘤、肾上腺疾病或其他疾病导致脏器功能衰竭者；
④葡萄糖-6-磷酸脱氧酶（G6PD）缺乏症患者；
⑤BMI＜18kg/m2者；
⑥近 3 个月使用过任何激素类药物及对胰岛素分泌有影响的药物者；
⑦对小檗碱过敏者，怀疑或确有酒精、药物滥用病史的患者；
⑧依从性差者。

Exclusion criteria：

1. Pregnant or lactating women;
2. Combined with cardiovascular diseases, liver and kidney and hematopoietic system and other serious primary diseases, mental patients;
3. Combined with malignant tumors, adrenal diseases or other diseases leading to organ failure;
4. Patients with glucose-6-phosphate deoxygenase (G6PD) deficiency;
5. BMI <18kg / m2;
6. Anyone who has used any hormonal drugs and drugs that have an effect on insulin secretion in the past 3 months;
7. Those who are allergic to berberine, who are suspected or have a history of alcohol or drug abuse;
8. People with poor compliance.
