研究目的:

采用低深度全基因组测序(sWGS)检测技术,检测200例初诊AML和MDS患者的全基因组序列,分析全基因组范围内的大片段缺失与重复,探索低深度全基因组测序(sWGS)检测CNA与核型分析、荧光原位杂交检测的一致性,分析sWGS对CNA的高检出率对疾病预后判断的指导意义。  

Objectives of Study:

Shallow whole-genome sequencing (sWGS) was utilized to depict genomic CNA profiles from the bone marrow cells of 200 newly diagnosed patients with AML/MDS. The large-scale deletions and duplications across the entire genome was analyzed to explore the concordance of genomic abnormalities identified by sWGS with those identified by cytogenetics and FISH. The objective of the study is to evaluate the clinical utility of sWGS based CNA analysis in myeloid malignancies.

纳入标准:

(1)根据2016 WHO诊断标准诊断初诊AML、MDS患者或MDS继发AML患者
(2)18≤年龄≤75,性别不限
(3)AML患者需进行染色体核型分析检测
(4)MDS患者需进行染色体核型分析检测、荧光原位杂交FISH(-5/5q-、-7/7q-、+8、20q-、-Y(男性测,女性不测))检测
(5)体力评分 0-3 级(WHO 标准)

Inclusion criteria

(1) Newly diagnosed AML, MDS and secondary AML patients according to 2016 WHO diagnostic criteria
(2) Aged 18 to 65 years old, male or female
(3) AML patients who have undergone cytogenetics
(4) MDS patients who have undergone cytogenetics and FISH (-5/5q-, -7/7q-, +8, 20q-, -Y (male test, female excluded))
(5) Performance status 0-3 (WHO standard)

排除标准:

(1)AML患者未进行染色体核型分析检测
(2)年龄≤18或年龄≥75岁
(3)MDS患者未进行染色体核型分析检测、荧光原位杂交FISH(-5/5q-、-7/7q-、+8、20q-、-Y(仅限男性))检测
(4)同时患有其它脏器恶性肿瘤
(5)存在其他阻碍研究进行的各类情况

Exclusion criteria:

(1) AML patients who have not undergone cytogenetics
(2) Age ≤ 18 or age ≥ 75 years old
(3) MDS patients who have not undergone cytogenetics and FISH (-5/5q-, -7/7q-, +8, 20q-, -Y (male only))
(4) having malignancies other than myeloid tumors
(5) Other conditions that deemed to be excluded from the study at the investigator's discretion.
