研究目的：

主要目的：通过多中心随机对照临床试验研究,探讨诊断超声联合微泡增强晚期胰腺癌化疗的安全性和有效性。 次要目的：评价诊断超声联合微泡增强临床胰腺癌化疗能够提高晚期胰 腺癌患者的总体生存期、良好生存期 (PS<2分 )和疾病控制率 (DCR)的能力。  

Objectives of Study：

Objective: To investigate the safety and efficacy of ultrasound combined with microbubble enhanced chemotherapy for advanced pancreatic cancer. Secondary objective: To evaluate the ability of diagnostic ultrasound combined with microbubble enhanced clinical chemotherapy in improving the overall survival, good survival (Ps < 2) and disease control rate (DCR) of patients with advanced pancreatic cancer.

药物成份或治疗方案详述：

 

Description for medicine or protocol of treatment in detail：

 

纳入标准：

① 经病理组织学或细胞学证实为无法手术切除的胰腺癌局部晚期 (Ⅲ期)伴有或不伴有远处转移 (Ⅳ期)患者,不论转移灶数量多少；
② 年龄 18-75岁 ,男女不限；
③ 肿瘤学评估(超声、CT或 MRI)有可测量和可评价的肿瘤病灶；
④ 预计生存期 3个月以上;PS评分 0-l分；
⑤ WBC≥3.5X10^9/L、 ANC≥1.5X10^9/L、PLT≥80X10^9/L、Hb≥90/L；
⑥ 肝肾功能无明显异常: ALT、 AST≤正常值上限2.5倍 ,Urea、 Cr≤正常值 上限1,25倍 ,肝转移患者ALT、 AST≤正常值上限5倍; ⑦心肺功能基本正常；
⑧ 无其他恶性肿瘤病史；
⑨ 依从性良好,无精神障碍。

Inclusion criteria

1. The patients with locally advanced pancreatic cancer (stage III) with or without distant metastasis (stage IV); confirmed by histopathology or cytology were unresectable, regardless of the number of metastases;
2. Age 18-75 years old, male or female;
3. Oncological evaluation (ultrasound, CT or MRI) showed measurable and evaluable tumor lesions;
4. The estimated survival time was more than 3 months;
5. WBC >= 3.5x 10^9/L, ANC >= 1.5 x 10^9/L, PLT >= 80 x 10^9/L, Hb>= 90/L;
6. There was no significant abnormality in liver and kidney function: ALT and AST <= 2.5 times of the upper limit of normal value, urea and Cr <= 1,25 times of the upper limit of normal value, ALT and AST <= 5 times of the upper limit of normal value in patients with liver metastasis;
7. Cardiopulmonary function was basically normal;
8. There was no history of other malignant tumors;
9. Good compliance, no mental disorder.

排除标准：

① 妊娠或哺乳；
② 对化疗药物过敏及超敏体质；
③ 肠梗阻或不全性肠梗阻；
④ 伴有大量腹水；
⑤ 伴有脑、骨转移；
⑥ 正在接受其他抗肿瘤药物治疗或放疗；
⑦ 同时参加其他临床试验或化疗结束时间小于4周。

Exclusion criteria：

1. Pregnancy or lactation;
2. Allergic to chemotherapeutic drugs and hypersensitivity constitution;
3. Intestinal obstruction or incomplete intestinal obstruction;
4. There was a large amount of ascites;
5. With brain and bone metastasis;
6. Receiving other anti-tumor drugs or radiotherapy;
7. At the same time to participate in other clinical trials or chemotherapy end time is less than 4 weeks.
