本课题主要研究组织干细胞恶变为 CSC 的调控机制以及组织干细胞变异、恶变为CSC 的早期识别及基于组织干细胞变异和 CSC 的诊治新策略。鉴定导致组织干细胞形成 CSC 的关键分子;找到导致肿瘤发生过程中 CSC 异质性产生的关键性乙酰化调控因子;探讨滤泡辅助 T 细胞( TFH)与 CSC 的相互作用,探讨 CSC 免疫逃逸机制和治疗意义;明确上述关键分子在早期诊断、精准治疗选择、疗效评估和预后判断中的意义;应用规范和标准化的组织干细胞、CSC 和病理组织样本库(含血液)以及临床信息资源,开展回顾性研究,探讨本项目研究成果在组织干细胞应用中的变异评估、恶性肿瘤早期诊断、精准治疗选择、疗效评估和预后判断中的意义。  

Objectives of Study:

This program mainly studies the mechanism of CSC formation from tissue stem cell, the early identification of the malignant transformation of tissue stem cell, and the new diagnostic and therapeutic strategies based on CSC formation from tissue stem cell. The detail objectives are as follows: (1) to identify the key molecules that regulate the CSC formation from tissue stem cells; (2) to find key acetylation regulators for CSC heterogeneity during tumorigogenesis; (3) to explore the interaction of follicular-assisted T cells (TFH) and CSC, and thus to explore the mechanism of immune escape of CSC and its therapeutic significance; (4) to clarify the significance of these key molecules in early diagnosis, precise treatment selection, efficacy evaluation, and prognostic judgment; (5) to explore the significance of the research results in the evaluation of tissue stem cell variation, early diagnosis of the malignant tumors, precise treatment selection, efficacy assessment, and prognostic judgment, by using standardized tissue stem cells, CSC, pathological tissue sample database (including blood), and clinical information resources, to carry out retrospective research.

纳入标准:

(1)具有手术切除或穿刺组织的胶质瘤病例;
(2)PDGFRA和P53的阳性染色;
(3)患者手术前未进行放疗或化疗。

Inclusion criteria

(1) The glioma samples are from surgical resection or puncture;
(2) The samples are positive staining for PDGFRA and P53;
(3) The patients have no history of radiotherapy or chemotherapy before surgery.

排除标准:

(1)肿瘤级别未定和诊断不明确的病例。
(2)患有传染性疾病的病例。
(3)排除研究人员认为其他原因不适合的病例。

Exclusion criteria:

(1) Cases with uncertain tumor grade and unclear diagnosis.
(2) Cases with infectious diseases.
(3) Other cases unsuitable for the study confirmed by the researchers.
