研究目的：

多囊卵巢综合征(PCOS)与胰岛素抵抗(IR)密切相关。骨形态发生蛋白-9 (BMP-9)在维持葡萄糖稳态中发挥重要作用，但BMP-9与多囊卵巢综合征之间的关系尚未见报道。在此，我们报道BMP-9在多囊卵巢综合征中的变化及其对IR的影响。  

Objectives of Study：

Polycystic ovary syndrome (PCOS) is closely related to insulin resistance (IR). Bone morphogenetic protein-9 (BMP-9) plays an important role in maintaining glucose homeostasis, but an association between BMP-9 and PCOS has not been reported. Here, we report the changes in BMP-9 and the influence of this protein on IR in PCOS.

药物成份或治疗方案详述：

PCOS患者中有25例接受艾塞那肽干预治疗，第1个月为5μg/d，第2、3个月为10μg/d。GLP-1给药剂量按用药说明书选择。 

Description for medicine or protocol of treatment in detail：

Among the PCOS patients, 25 received intervention treatment with EX at 5 μg/d in the 1st month and 10 μg/d in the 2nd and 3rd months. GLP-1 administered doses were selected according medicine instruction. 

纳入标准：

PCOS的诊断依据2003年欧洲鹿特丹人类生殖与胚胎学会和美国生殖医学学会(ESHRE/ASRM)的诊断标准，其中规定诊断需要以下3项中的任意2项:
1.罕见排卵和/或无排卵；
2.临床表现和/或雄激素过多；
3.超声检测卵巢多囊性改变，即一个或两个卵巢内>=12个直径2- 9mm的卵泡，/或卵巢体积>=10 ml。所有患者均采用正血糖高胰岛素钳夹(EHC)技术评估；M值(葡萄糖代谢率)小于6.286 mg/kg/min表示IR, M值大于或等于6.286 mg/kg/min表示非IR。其他已知内分泌系统相关性腺疾病(如先天性肾上腺增生、雄激素分泌瘤和库欣综合征)的患者被排除在外。

Inclusion criteria

PCOS was diagnosed according to the 2003 European Society for Human Reproduction and Embryology of Rotterdam and the American Society for Reproductive Medicine (ESHRE/ASRM) diagnostic criteria, which stipulates that any two of the following 3 items are required for a diagnosis:
1.Rare ovulation and/or anovulation;
2.Clinical manifestations and/or hyperandrogenism;
3.Ultrasound detection of polycystic changes in the ovaries, i.e., >=12 follicles of 2-9 mm in diameter in one or both ovaries and/or ovarian volume >=10 ml. All patients were evaluated using the euglycaemic hyperinsulinaemic clamp (EHC) technique; an M value (glucose metabolic rate) less than 6.286 mg/kg/min indicated IR, and an M value greater than or equal to 6.286 mg/kg/min indicated non-IR. Patients with other known endocrine system-related gonadal diseases (such as congenital adrenal hyperplasia, an androgen-secreting tumour and Cushings syndrome) were excluded.

排除标准：

1.年龄大于40岁;
2.体重指数(BMI)大于35 kg/m^2；
3.有其他导致不孕的疾病的；
4.有明显的肝肾疾病体征或症状，谷丙转氨酶高于正常值的2倍，血清肌酐高于正常值，严重的胆结石，甘油三酯高于5mmol /L，心力衰竭，甲状腺疾病，或有糖尿病史；
5.最近3个月内使用影响糖脂代谢的药物；
6.近3年有精神障碍或酗酒、吸毒史；
7.对研究药物(艾塞那肽(EX)注射液)有过敏史或禁忌症；
8.尿液HCG阳性或在研究过程中不愿意采取严格的避孕措施；
9.参与其他药物试验。

Exclusion criteria：

1.Older than 40 years;
2.Body mass index (BMI) greater than 35 kg/m^2;
3.The presence of other diseases that cause infertility;
4.Obvious signs or symptoms of liver or kidney disease, ALT higher than 2 times the upper limit of normal, serum creatinine level above normal, severe gallstones, triglycerides greater than 5 mmol/L, congestive heart failure, thyroid disease, or a history of diabetes;
5.The use of drugs that affect glycolipid metabolism within the previous 3 months;
6.A mental disorder or a history of alcohol or drug abuse in the past 3 years;
7.A history of allergies or contraindications to the study drug [exenatide (EX) injection];
8.A positive urine HCG test or unwillingness to follow strict contraceptive measures during the study; and
9.Participation in other drug trials.

非项目组成人员采用分层随机方法，入组时将受试者分为PCOS组、IR组、NC组，每层均采用简单随机法，通过随机数字产生随机数，按照随机数分为试验组和对照组。
