研究目的：

通过病例-对照研究，探讨膳食肌醇六磷酸（IP6）的摄入与结直肠癌的患病风险性，同时采用肠道菌群分离鉴定和肠道菌群16s宏基因组测序分析方法，分析结直肠癌患者肠道菌群构成，探讨人体肠道菌群及细菌产植酸酶活力与结直肠癌患病风险性的关系。  

Objectives of Study：

We will determine the relationship between IP6 intake and the risk of colorectal cancer (CRC) in China through case - control study, explore the composition change of human gut microbiota and contribution of change in gut microbiota and the microbial phytase activity variance to the risk of CRC by 16s rRNA metagenomic sequencing and traditional culture techniques.

纳入标准：

病例（结直肠癌）组：
1、经乙状结肠镜和(或)纤维结肠镜确诊的门诊首发结直肠癌患者；
2、年龄在30-70岁之间的男、女患者；
3、既往无结肠或直肠手术史，无肠道肿瘤、炎性肠病或炎症病史；
4、无家族性腺瘤性息肉病或遗传性非息肉病性肠癌家族史；
5、具有配合研究者进行调查研究的认知和行为能力；
6、能够提供签署的书面知情同意书；
7、能够向研究者提供有关具体食物和食物种类以及微量元素摄入等相关详细资料信息；
8、愿意提供2份正常排出的新鲜粪便标本(粪便采集时间在肠镜检查完毕后2-4周，且在手术清洁灌肠和应用口服抗生素 之前)。
对照组：
1、无肠道疾病症状或病史(如结肠腺瘤性息肉，结直肠癌或炎症性肠病)的健康志愿者和同期住院的创伤患者；
2、年龄和性别与病例组相匹配；
3、在一级亲属中无结直肠癌患者；
4、体重指数在20-28kg/m^2之间；
5、能够提供签署的书面知情同意书；
6、能够向研究者提供有关具体食物和食物种类以及微量元素摄入等相关详细资料信息；
7、愿意提供2份正常排出的新鲜粪便标本。

Inclusion criteria

For case group:
1．Consecutive outpatients suffering from a newly diagnosed colorectal cancer(CRC) by colonoscopy and 　 pathology;
2．Both males and females, aged 30-70 years;
3．No previous history of colon or rectal surgery, of diseases such as cancer, or of inflammatory or infectious injuries of the intestine;
4．None familial adenomatous polyposis or hereditary nonpolyposis colon cancer;
5．Have mental competency to complete the interview;
6．All individuals will provide written informed consent prior to participating in the study;
7．A detailed information on frequency of consumption of specific foods, food groups, and micronutrients, will be collected by interview;
8．Willing to collect 2 spontaneously passed fresh stool samples between 2 and 4 weeks after the diagnostic colonoscopy and biopsies, and before starting pre-operative preparation with bowel cleansing and with oral antibiotics.
For control group:
1．Clinically healthy volunteers with no symptoms or history of intestinal disease(e.g.colonic adenomatous polyps, CRC or inflammatory bowel disease);
2．Being matched for age and sex to the case group;
3．No family history of CRC in first-degree relative;
4．Body mass index(BMI) between 20 and 28 kg/m^2;
5．Subjects giving his/her written informed consent;
6．A detailed information on frequency of consumption of specific foods, food groups, and micronutrients, will be collected by interview;
7．Willing to collect 2 spontaneously passed fresh stool samples.

排除标准：

1、孕妇或哺乳期妇女；
2、不能提供签署的书面知情同意书；
3、取粪便标本前1个月内应用过抗生素；
4、取粪便标本前1个月内应用过促胃肠动力药和包括酸奶在内的益生菌饮料；
5、规律服用阿司匹林和(或)其他非甾体类消炎药；
6、在取粪便标本前进行过化疗或放疗；
7、目前患有肠道致病菌感染；
8、1个月内出现明显的胃肠道出血(包括怀疑因痔疮导致的出血)；
9、慢性腹泻、炎性肠病、复发结直肠癌、肠易激惹综合征、其它胃肠道疾病、胃肠道手术(如结肠切除术、直肠切除术 、胆囊切除术)以及其它慢性疾病(如严重脏器衰竭、其它部位的恶性肿瘤、糖尿病)；
10、体重指数≥28kg/m^2；
11、患者肠镜检查完毕后到进行手术之间的间隔时间小于2周。

Exclusion criteria：

1．Pregnant or lactating women;
2．Inability to provide informed consent;
3．The use of antibiotics within 1 month before stool sampling;
4．The use of prokinetic drugs and probiotics including yoghurt within 1 month before stool sampling;
5．Regular use of aspirin and/or nonsteroidal anti-inflammatory drugs;
6．Undergone chemotherapy or radiation treatments before sampling;
7．Current infection with an enteric pathogen;
8．Overt gastrointestinal bleeding within 1 month (including due to suspected hemorrhoids);
9．Chronic diarrhea, inflammatory bowel disease, recurrent colorectal cancer, irritable bowel syndrome, or other gastrointestinal disorder, gastrointestinal surgery (e.g.colectomy, proctectomy or cholecystectomy ), severe chronic diseases such as major organ failure,other malignant diseases,diabetes mellitus;
10．Body mass index(BMI) >= 28kg/m^2;
11．Time interval between colonoscopic biopsy and operation less than two weeks.
