研究目的: |
肺癌是我国发病率和死亡率最高的恶性肿瘤之一,小细胞肺癌 ( small cell lung cancer,SCLC) 约占肺癌的20%,恶性程度高、预后差。且目前尚无有效的分子靶向药物。早期诊断及早期治疗能大大改善预后。因此,探讨其发病的分子机制,寻找早期诊断方法,是目前SCLC研究的主要方向。 近年来,肿瘤标志物的识别已成为恶性肿瘤早期诊断的研究热点。肿瘤标志物是指在肿瘤发生和增生过程中,由肿瘤细胞生物合成、释放或者是肿瘤与宿主相互作用而产生,并能反映肿瘤发生、发展的一类代谢物质。传统肿瘤标记物如神经烯醇化酶对SCLC早期诊断的敏感度和特异度较低,难以达到早期准确诊断的目的。而近年快速发展的新一代代谢组学技术为发现和检测代谢肿瘤标志物提供了有力工具。该技术可以对人体肿瘤发生发展过程中的所有小分子代谢产物进行定性和定量分析,并能识别未知代谢产物。 本研究通过对SCLC患者血样以及细胞的代谢组学差异分析,找到SCLC可能的肿瘤标志物,并对其进行鉴定和功能验证,为进一步研究SCLC发病的分子机制、开发靶向治疗药物以及针对病人个体的精准医疗奠定基础。 |

Objectives of Study: |
Lung cancer is one of the malignant tumors with highest morbidity and mortality in China. Small cell lung cancer (SCLC) accounts for about 20% of lung cancer, with high malignancy and poor prognosis. And there is no effective molecular targeting drug. Early diagnosis and early treatment can greatly improve the prognosis. Therefore, to explore the molecular mechanism of its onset, to find early diagnostic methods, is the main direction of current SCLC research. In recent years, the identification of tumor markers has become a hot topic in the early diagnosis of malignant tumors. Tumor markers refer to a class of metabolites that are produced by tumor cell biosynthesis and release, or by tumor-to-host interactions during tumorigenesis and hyperplasia, and can reflect tumorigenesis and development. Traditional tumor markers such as Neuron specific enolase have a low sensitivity and specificity to early diagnosis of SCLC, and it is difficult to achieve the purpose of early accurate diagnosis. In recent years, the rapid development of a new generation of metabolomics technology provide a powerful tool for the discovery and detection of metabolic tumor markers. This technique allows qualitative and quantitative analysis of all small molecule metabolites in the development of human tumors and identifies unknown metabolites. In this study, the SCLC tumor biomarkers will be identified by metabolomic analysis of SCLC patients and cells, and its function will also be verified. Our work will lay the foundation of the molecular mechanism of SCLC pathogenesis, the development of targeted drugs and the precision medical care for SCLC patients. |
